Zosuquidar trihydrochloride (LY335979) is a third generation modulating agent that was developed specifically as a selective, MDR1/P-glycoprotein (P-gp) inhibitor. In vitro concentrations of zosuquidar from 50 to 100 nm are capable of circumventing P-gp-mediated drug resistance in virtually every cell culture system tested. In experiments using tumor-bearing mice with syngeneic and human xenografts, zosuquidar was efficacious in restoring drug sensitivity to P-gp expressing implants. Moreover, preclinical studies using murine and canine models demonstrated that zosuquidar had no observed effect on the pharmacokinetic profile of coadministered P-gp substrates, including doxorubicin and paclitaxel. These data suggest that zosuquidar warrants investigation in clinical trials aimed at reversing drug resistance mediated by P-gp.
